I honestly find it very unlikely that any of the others would easily come into conflict with Twilight now. I think with her being a Princess and all that they would be more likely to defer to her decisions. Might actually be an upcoming episode dealing with that sort of thing.
Now that you mention it, I really want to see season 4 address this. Maybe even make it a subplot across several episodes.
Twilight could want everyone to treat her exactly the same—she has to learn that things can’t go back to exactly how they were before, that she has authority now and everyone’s depending on her to use that authority. On the other hand, the citizens of Ponyville would go too far in deferring to Twilight and make her uncomfortable with all their grovelling, so they’d need to be reminded that Twilight’s the same pony as before under the tiara.
I’d like to see a scene like, say, Twilight participating in some event on the scale of Winter Wrap Up, and very pointedly deferring to Mayor Mare’s authority.

I'd definitely love to see the awkwardness of everypony learning to get used to Twi getting her Class Promotion. Twi definitely will be insisting ponies don't have to be so stiff and formal around her lol, she'll know what it's been like for Celestia all along.
