Celestia will give Ponyville to Twilight to rule. In conjunction with having given Cadance a chunk of land up north to rule, the next series antagonists will be the current nobility worried that Celestia is going to give away their land chunk by chunk to ponies easier to control.

To do that, she'll need to build an observatory, get some Great Scientists to build a couple academies, and try to rush stuff like the Porcelain Tower or the Great Library.
The Great Library is useless. What you really want are stuff like Darwin's Voyage and SETI. Of course the real way to get uberfast research is to smallpox across a continent with deomcracy and 0% tax.

Twilight would be a competent ruler as long as she delegates and doesn't try to micro-manage. Winter Wrap Up proves that she's good with organization, so she probably has that side of things well under wraps. Most of her problems would stem from the diplomacy/PR side of things. I could easily see her balancing the budget or writing legislation or whatever, but dealing with ambassadors and lobbyists and stuff would be a problem.
