I'd always taken both the Royal Canterlot Voice and the archaic mannerisms as a sort of front. She drops it completely when she's more comfortable, she just uses them when adressing her subjects. For example, we see no hint of either when she's apologizing to her sister in episode 2.
I always imagine Twilight as black myself, and RD as Dominican.
I tend to think Pinkie is a bit more well rounded then the rest, but by no means fat, or even chubby, just, she's got a bit of padding. I also bet her weight looks like a roller coaster depending on the sugar/hyperness ratio for the last week or so.

I also prefer women with a bit of meat on their bones. My problem comes when people start taking that phrase figuratively, because then I have to explain 'like an average person and not twenty to thirty pounds overweight'. =|
I've got nothing against you as an artist, Mousa, but your design of Pinkie's just flat-out beyond my limits of attractiveness. I realize others disagree, and all the better; but I'll continue to visualize the girls in the way that's most aesthetically and characteristically pleasing to me.

As far as ethnicty, if we're not just going with the default, all white cast.
Twilight I'd see as Asain, or maybe Indian (India type not Native American type)
Rarity as Jewish
RD as either black or latino
AJ Could be either Black or White southerner.
pinkie could be anything
And Fluttershy seems the most assuredly white.
