Twilight was applying for a school, right? So if it began in elementary school, would that mean she was 5 going on 6 when she got her cutie mark? Meaning she's 5 years older than Spike...?

we know for a fact that she was old enough to read quite well at the time - burning through several sizeable and likely very complex spellbooks, so she likely would have been in school for a while. She was also at the right age to get her cutie mark - if the 'puberty metaphore' theory is too believed, then we can assume she was likely in the range of 8-10...for how excited she seemed about that part especially, I'm going to assume it's on the long end of the scale. Faust says that The Mane cast is about 16-17 years old (though again, twilight is a grad student, living a good ways from home, so I'm going to go for 18-19), which would put Spike at about 7-9. Which seems about right, given his behaviour, really.
Besides, it's entirely possible that ponies mature in a different rate than humans.

And then I can't help but wonder how much older Rarity is than him. Would Rarity be older or about the same age as Twilight? I kind of always thought of the Spike/Rarity relationship like the Finn/Princess Bubblegum relationship. I think it's adorable, but wouldn't Spike be like 10 years younger than her?
I think Fluttershy's size compared to the others in that episode was because she was a year older. If that's the case, it seems as if ponies mature a bit faster.
We've also seen some background ponies that appeared to be the same age, so maybe they don't age very fast after reaching adulthood. Plus, Granny Smith looked just as old, just without her walker and possibly less senile.

So I think I might actually be willing to give this show a shot.... assuming I can do it a manner that ensures that no one I know will ever find out.

I'm a sucker for cartoons that actually have decent characterization.

Do go ahead. Have a bit of patience especially if you're starting off from the beginning, as the pilot is more typical of what you'd expect an MLP show to be like, although it's still entertaining. You could look at ones like the Ticket Master, Suited For Success, Over a Barrel, and I think Swarm of the Century is nice as well. The newest episode requires some background, mostly the episodes Sonic Rainboom and Call of The Cutie, the former being one of the better episodes and the latter being bit of a YMMV but as the rest of the series, still fun to watch.
