So I keep trying to imagine Pinkie Pie as a human.
Not so much what she would look like, but how she acts. For some reason the idea of a human acting like Pinkie amuses me.

Do you mean what she would be like as a real human in the real world? I imagine the 4th wall antics wouldn't be a part of her repertoire anymore.

I can't help but think her activities might not stay so G-rated. I bet she'd be fine, but I bet a lot of what makes her an entertaining character would grate on your nerves in real life.
And I'm betting Dash would be in trouble without the cartoon physics safety net. Fluttershy would probably be even more of a neurotic mess having knowledge of all the dark crap in the world. Applejack could do okay, but I'm not so sure she'd be as thrilled about managing the kinds of farms we have; I guess there'd be less opportunities to work with your hands.
Rarity and Twilight would probably be pretty happy, though. Their skills and interests line up pretty well with what's in demand in the economy.

Picking apples by kicking them out of trees sounds significantly more badass than the normal way.

I think Fluttershy would be just fine. The Real World isn't that dark. She may or may not be a tree hugger. And if someone gives her trouble, she stares them down with a fierce gaze. Rainbow Dash would be an adrenalin junkie and probably prone to trying daring things...also part time pilot. And Pinkie Pie would probably be that one person who pretty much pops up where you least expect them too...like in a fruit stand. And she would be aware if some random stranger is looking her way while she is talking with a friend and give a glance. meanwhile, Lyra and Bonbon would be a lesbian couple battling diabetes and over-eating. And Derpy would be a loving mother and hard working postal worker who gets the job done despite a vision problem.
Oh, and Pinkie Pie would also be the kind of person who can organize huge public events at a moments notice.
Applejack could probably take her love of farming and turn it into a big career by developing more efficient farming techniques and such.

Also, Fluttershy would be able to get help from a Therapist who could help her get past that shyness problem if she was really that incapable of operating in the real world.

I don't think Fluttershy would need a therapist seeing as she'll still be friends with the others. We are working under the assumption that the friends are still friends and not separated, right?
I wonder if it would be appropriate to make "Buck Norris" jokes.
Anyway, Fluttershy. Sure, she has a bit of dracophobia, and can be a something of a pushover, and she's plenty shy, but not cripplingly so, in my opinion.
I mean, if she can talk down the object of her worst fears...
Come to think of it, she managed to pull that off, she has "the stare"... maybe her shyness is just a way of holding back anger or frustration? Something like that? Hmm...

Fluttershy's shyness isn't a personality quirk. It's a safely mechanism. Princess Celestia put it there to protect Equestria from 'the Stare'.
Yeah. You know how Australia doesn't have an ocean to keep people out, but rather to keep the wildlife in? It's kind of like that.

I wouldn't mind someone like Pinkie Pie in real life As for the "not so G-rated antics", I'm thinking of giving her a beer bottle if I ever draw the ponies in human form.
