RD seems more like she's the one that gets things done, not the leader. Now Applejack, she seems like good leader material in case Twilight ever gets incapacitated or something.

RD is far too self-absorbed to be an effective leader.
She's not my favourite pony, but I think that Rarity is the one who is the most together of the Mane 6. She's a bit (OK, a
lot) of a drama queen, but she has both organizational talent and a good grasp of people skills (OK, pony skills).
Applejack is a cool, but she's a bit too narrow in her outlook to be a good leader. Fluttershy would go catatonic. And Pinkie Pie, of course, would make the
others go catatonic.
But they seem to

Now I'm imagining Pinkie Pie as a Quest Giver:
