For me, it's asexual until proven otherwise. And so far, only Rarity has settled on a side.

Here's how I break it down:
- Applejack isn't interested in anypony unless her love life is very very private.
- Rarity is Camp Straight.
- Rainbow Dash is theoretically open to relationships of either sex, but is so focused on flying and competition it's rarely crossed her mind. Though I suspect there's some repressed sexuality in her affection for the Wonderbolts.
- Fluttershy is asexual for the most part aside from a one-sided crush on Rainbow Dash.
- Pinkie doesn't see herself as anything other than what she is. To her, fun is fun no matter the gender, the concept probably never crossed her mind. She'd probably be confused if someone ever explained the concepts of orientation to her.
- I can't see Twilight as anything else but into mares. All meaningful relationships she's ever had have been with girls and arguably Spike and Shining Armor are the only strong bonds she has with males in her life. I can't be sure if she would be into stallions, but she's been around females so much it's easier for her to form attachments to them.
Though it would take quite the pony to get her affections away from books or her friends long enough to maintain an interest.

@Story I like to think that Scootaloo's parents are actually really worried about her, and that while they know Dash is alright, they're still wondering why Scoots likes Dash more than them.

I enjoy every damn second of it!
I don't like the idea of basing Twilight's sexual orientaion on the meaningful relationships she's had recently. Until episode 1, it seems she had almost no real meaningful relationships (or, at least, she thinks she didn't). Her relationships in Ponyville are ones of friendship, not romance.
