If you want a spoilery explanation now:
the Planet Express crew accidentally time-travels back to Roswell, NM on July 7, 1947, causing the Roswell Incident. Along the way, Philip J. Fry meets Enos Fry, the man he believes to be his grandfather, who has yet to conceive Philip's father. Philip J. Fry's attempts to protect Enos end up killing him instead. Fry wonders why the heck he doesn't cease to exist now that his grandfather is prematurely dead, and he comes to the conclusion that Enos wasn't really his grandfather, and Eno's girlfriend Mildred wasn't really his grandmother. So when Fry's attempt to console Mildred result in Mildred offering to sleep with him, Fry takes her up on the offer. The morning after, the rest of the Planet Express crew inform Fry that Mildred really is Fry's grandmother—by sleeping with her, he just became his own grandfather.
This event actually becomes important later: Because of his unique ancestry, Fry is a genetic anomaly, which makes him the only human in the universe immune to the Brainspawns' attacks.
