I don't think leadership is something that stresses Twilight out at all. She's really on top of things whenever it comes to that. Really, it's mostly trying to impress Celestia that stresses her out.
I mean apart from Discord and the Changelings and Appleloosa, there aren't many Incidents that we get to know of outside Ponyville.

I think as long as Twi had someone to help keep her grounded, she'd make a fine mayor. Her biggest points of vulnerability tend to be when she's stuck in a tough spot all on her own.
