Yep, being on Skype and watching Bleach will slow Defrag down. The problem is that Defrag is having to find a way to move those bytes without messing up an active process. Imagine trying to rearrange a room while two kids are in it watching a movie, and you're trying very hard not to interrupt them, and you have an idea what Defrag is going thru.
I personally defrag once a month, and always at a time when I know I won't be using the computer for awhile. And I run Spybot, Malwarebytes and Avira Antivirus scans every night (those
can be run while you're doing other things). Windows 7 works fine for me.
Edit - And yes, Defrag ASAP and on a regular basis. I had a computer I used all the time but never defragged for two whole years. By that time, the hard drive was so effed up that defrag would always just cop out. And this severely hampers performance.
On-Topic Edit - Personally I imagine the ponies being Windows-users, though it wouldn't surprise me if Twilight Sparkle (and maybe Pinkie Pie) had some programming/hacking skills.
