I'm sure she reads every note Twilight sends her. It's clear (
Alternative Character Interpretation not withstanding) that she cares about Twilight, and the whole "learn about friendship and report to me on your findings" is for Twilight's benefit, not Celetia's. Celestia knew Twilight well enough to know making it a school assignment was the best way to make sure she took it seriously and gave it her all. Celestia doesn't read them like a teacher grading her student, more like a family member reading a letter about how Twilight's been doing.
